What Is Palmitoyl Tripeptide-5?
This is a synthetic peptide which mimics the sequence of the protein Thrombospondin I (TSP-1) fragment, presenting the sequence N-(1-oxohexadecyl)-L-lysyl-L-valyl-L-lysine.
Initially, this tripeptide was produced with the help of solid-phase synthesis. But, since this mode of production caused significant cost and pollution to the environment, two different liquid methodologies were introduced.
What Does It Do?
Palmitoyl Tripeptide-5 was hypothesized to reduce metalloproteases (MMP’s) expression and pro-inflammatory cytokine synthesis; these effects result in vasodilation and increased capillary permeability.
This is why this cosmetic peptide was first used as an ingredient in products designed to reduce skin redness in type I rosacea. It was also used in anti aging cosmetic products, formulas for sensitive skin, as a type I and II collagen synthesis promoter, as well as a melanin production inhibitor (aimed at reducing tyrosinase activity).
How Effective Was Palmitoyl Tripeptide-5?
As mentioned through the text, for some of these studies we only have the basic information and can not conclusively say how effective this tripeptide really was. Luckily, we have one animal study done in vitro, that showed Palmitoyl Tripeptide-5 therapy significantly decreased collagen breakdown process.

SNAP-8 is a mimetic of the N-terminal end of SNAP-25 (SyNaptosome-Associated Protein). SNAP-8 competes with its 25 counterpart for a position within the SNARE complex and, by doing so, changes its formation. SNARE complex (SNAP REceptors complex) primary role is to mediate the fusion of vesicles with the target membrane. Some of these vesicles contain neurotransmitters responsible for botulism and tetanus, suggesting that the SNARE complex affects muscle contractions and prevents fine lines from being formed.
SNAP-8 is a synthetic peptide that mimics the effects of botulinum. Unlike botulinum which stops the release of chemical acetylcholine (which makes the muscles contract), SNAP-8 disrupts the SNARE complex. This will, then, prevent the release of acetylcholine and prompt temporary muscle relaxation.

Pentapeptide-18 belongs to the group of neurotransmitter peptides and is associated with the SNAP-25 receptor protein (SyNaptosome-Associated Protein, 25 kDa). This receptor protein is involved in the release of acetylcholine and the formation of SNARE (SNAP receptors) complex.
Pentapeptide-18 affects the neuromuscular synapses and, by binding to these synapses, it closes up the calcium channels, which results in the reduction of acetylcholine release. Acetylcholine is responsible for muscle contractions and the formation of wrinkles; so, by blocking it, researchers believe they can affect the formation and the severity of wrinkles.
